Intracellular flow cytometric analysis of4% paraformaldehyde-fixed, 90% methanol-permeabilized Daudi (human Burkitt's lymphoma cell line) cells labeling [MEF2B with ab242058 at 1/500 (red) compared with a rabbit monoclonal IgG (ab172730, black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti-Rabbit IgG H&L (Alexa Fluor^® 488) (ab150077), at 1/2000 dilution was used as the secondary antibody.

Immunoprecipitation - Anti-MEF2B antibody [EPR22193-26] - BSA and Azide free (AB242347)

MEF2B was immunoprecipitated from 0.35 mg of Ramos (human Burkitt's lymphoma cell line) whole cell lysate with ab242058 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab242058 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.

Lane 1 : Ramos whole lysate 10 μg (Input).
Lane 2 : ab242058 IP in Ramos whole cell lysate (+).
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab242058 in Ramos whole cell lysate (-).

Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 1 second.

MEF2B also known as myocyte enhancer factor 2B plays an integral role in regulating gene expression. It is a transcription factor belonging to the MEF2 family which also includes MEF2A MEF2C and MEF2D. MEF2B has a molecular mass of approximately 50 kilodaltons. Researchers have found its expression mainly in lymphoid tissues but it is also present in other tissue types. Through its actions MEF2B influences various cellular processes by binding to DNA and facilitating transcriptional activation.

Biological function summary

MEF2B functions as a regulator of immune system development and differentiation. It acts at the level of transcription to determine the fate of cells by its interactions. MEF2B often partners with various cofactors to form a complex which enhances its ability to control gene expression. It is known to be highly involved in muscle development and cell proliferation underlining its diverse roles in cellular biology.

Pathways

MEF2B plays important roles in two key signaling networks: the MAPK signaling pathway and the calcium signaling pathway.
