MERTK and TYRO3 are receptor tyrosine kinases that belong to the TAM (TYRO3 AXL MERTK) family. MERTK is often referred to as c-MERTK and TYRO3 as RSE or Sky. MERTK has a molecular weight of approximately 110 kDa and TYRO3 weighs around 105 kDa. MERTK and TYRO3 are mostly found in the immune system particularly in macrophages dendritic cells and also in retinal pigment epithelial cells. These receptors are important for regulating cellular processes including phagocytosis immunological homeostasis and tissue repair.

Biological function summary

These proteins play roles beyond membrane receptors and are pivotal for immune regulation and maintaining cell health. MERTK pairs with ligands such as Gas6 and Protein S leading to receptor-autophosphorylation and activating downstream signaling pathways. TYRO3 also operates in conjunction with similar ligands and both receptors form part of a complex that helps in cellular homeostasis and inflammation control. They contribute to processes like the apoptotic cell clearance which helps in resolving inflammation.

Pathways

Both proteins participate in significant signaling axes such as the PI3K/AKT and MAPK pathways. These pathways are linked with cell survival proliferation and differentiation. MERTK activates these pathways upon binding with its ligands influencing immune functions and cellular repair. Alyso TYRO3 associates closely with Ras and Erk proteins which are central parts of these signaling cascades impacting cellular cycles and apoptosis.

Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to several ligands including LGALS3, TUB, TULP1 or GAS6. Regulates many physiological processes including cell survival, migration, differentiation, and phagocytosis of apoptotic cells (efferocytosis). Ligand binding at the cell surface induces autophosphorylation of MERTK on its intracellular domain that provides docking sites for downstream signaling molecules. Following activation by ligand, interacts with GRB2 or PLCG2 and induces phosphorylation of MAPK1, MAPK2, FAK/PTK2 or RAC1. MERTK signaling plays a role in various processes such as macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization and engulfment (PubMed : 32640697). Functions in the retinal pigment epithelium (RPE) as a regulator of rod outer segments fragments phagocytosis. Also plays an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response by activating STAT1, which selectively induces production of suppressors of cytokine signaling SOCS1 and SOCS3.
