MLH3 also known as MutL homolog 3 is a DNA mismatch repair protein part of the MutL family. It has a molecular mass of approximately 160 kilodaltons (kDa). This protein is mainly expressed in the testis and also found in various other tissues at lower levels. MLH3 plays a mechanical role in DNA repair through the correction of mismatches that occur during replication.
Biological function summary

MLH3 acts within DNA mismatch repair processes to maintain genomic stability. It forms heterodimeric complexes with MLH1 another MutL homolog which is essential for its DNA repair function. This complex carries out the recognition and repair of insertion-deletion loops and base-base mismatches in DNA therefore ensuring fidelity during cell division.

Pathways

MLH3 operates within the DNA damage response and repair pathways. It interacts significantly with the MLH1 protein to facilitate mismatch repair activities. Additionally MLH3 participates in the homologous recombination pathway working alongside proteins such as MSH2 to secure proper chromosome segregation during meiosis.

MLH3 mutations or dysfunctions have associations with Lynch syndrome a hereditary condition that increases cancer risk especially colorectal cancer. Alterations in MLH3 in combination with MLH1 mutations have implications for mismatch repair deficiencies observed in these disorders. Additionally variances in MLH3 contribute to the progression of endometrial cancer often linked with other mismatch repair proteins like MSH6 in the disease pathology.
