The Molybdenum cofactor sulfurase (MOCOS) also known as MCS is an important enzyme in sulfur transfer for the activation of molybdenum cofactor. MOCOS weighs approximately 88 kDa and is found in a variety of tissues including liver and kidney. It plays a critical role in the biochemical process of activating the molybdenum cofactor which is essential for the proper function of certain enzymes.
Biological function summary

MOCOS contributes significantly to a sulfurase complex that includes other enzymes responsible for molybdenum cofactor biosynthesis and activation. It specifically introduces sulfur into the cofactor which is important for the functionality of molybdenum-dependent enzymes like xanthine dehydrogenase and aldehyde oxidase. MOCOS's activity impacts not just individual enzyme functions but whole biochemical pathways governed by these enzymes.

Pathways

MOCOS's biological function integrates into the purine degradation pathway and the aldehyde detoxification pathway. The enzyme works collaboratively with xanthine dehydrogenase in purine metabolism enabling the conversion of hypoxanthine to xanthine and subsequently to uric acid. MOCOS deficiencies relate to xanthinuria Type II characterized by an excessive accumulation of xanthine due to impaired xanthine dehydrogenase activity. Individuals with dysfunctional MOCOS can experience xanthine oxidation defects leading to potential renal complications due to stone formation. This relates closely to aldehyde oxidase whose reduced activity might impact drug metabolism and increase susceptibility to certain aldehyde-induced cellular damages.

Sulfation of molybdenum is essential for xanthine dehydrogenase (XDH) and aldehyde oxidase (ADO) enzymes in which molybdenum cofactor is liganded by 1 oxygen and 1 sulfur atom in active form (PubMed : 34356852). In vitro, the C-terminal domain is able to reduce N-hydroxylated prodrugs, such as benzamidoxime (PubMed : 16973608).
