The metallothionein transcription factor 1 also known as MTF1 is a zinc finger protein known for binding to metal-responsive elements (MREs) in the promoter region of target genes. MTF1 has a molecular mass of approximately 80 kDa. It is widely expressed in various tissues with abundant presence in organs like the liver and kidney where it plays a role in the cellular response to metal ions. This transcription factor is critical in modulating genes involved in metal ion homeostasis and detoxification.
Biological function summary

MTF1 regulates the expression of genes imperative for cellular metabolism and protection against oxidative stress. MTF1 functions as a monomer but may also interact with other transcription factors and regulators to exert its effects. Through its activity MTF1 mitigates the cellular damage induced by excess metal ions by upregulating metallothionein genes which bind and sequester metal ions. This activity assists in maintaining metal ion balance within the cell and reducing metal-induced oxidative stress.

Pathways

MTF1 plays a significant role in copper and zinc ion homeostasis pathways. It interacts with proteins such as metallothioneins (MTs) and zinc transporter proteins to regulate intracellular metal concentration. MTF1 also participates in the oxidative stress response pathway where it collaborates with other transcription factors like Nrf2 to activate the transcription of antioxidant genes. This involvement positions MTF1 as an important player in cellular defense mechanisms against oxidative damage.

Abnormal MTF1 activity has connections to certain types of cancer and neurodegenerative conditions like Alzheimer's disease. In cancer altered MTF1 expression can contribute to tumor progression by disrupting metal ion regulation which in turn affects cell proliferation and survival. It is associated with proteins like p53 which have roles in cell cycle and apoptosis. In neurodegenerative diseases impaired MTF1 function may lead to metal ion imbalance exacerbating oxidative stress and neuronal damage often in conjunction with dysregulated expression of metallothioneins and other metal-handling proteins.
