Flow Cytometry - Anti-Muscarinic Acetylcholine Receptor M4/CHRM4 antibody [18C7.2] (AB77956)

Overlay histogram showing SH-SY5Y cells stained with ab77956 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab77956, 1μg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2μg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

The Muscarinic Acetylcholine Receptor M4 also known as M4 receptor or CHRM4 is a G protein-coupled receptor with a mass of about 60-70 kDa. It is part of the muscarinic receptor family which includes five subtypes. Expression of CHRM4 occurs broadly in the central nervous system particularly within the brain's striatum and cortex regions influencing neuronal activity. The M4 muscarinic receptor plays a critical role in modulating neurotransmission responding to the neurotransmitter acetylcholine.
Biological function summary

The M4 muscarinic receptor influences several important processes by coupling with G proteins primarily inhibiting adenylate cyclase activity which results in reduced cyclic AMP levels. The M4 receptor is not generally part of a larger protein complex; rather it functions through interaction with associated proteins like Gi/Go proteins.
