HeLa (Human epithelial cell line from cervix adenocarcinoma) cells labeling Myozenin 2 (green) using ab237053 at 1/100 dilution in ICC/IF, followed by an Alexa-Fluor®488-conjugated goat anti-rabbit IgG (H+L). Countersatined with DAPI (blue).

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.

Paraffin-embedded human heart tissue stained for Myozenin 2 using ab237053 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Myozenin 2 also known as calsarcin-1 mechanically functions as a scaffold protein that anchors filamentous actin to the sarcomeric Z-disc in striated muscles. It weighs approximately 32 kDa. Myozenin 2 is primarily expressed in cardiac and skeletal muscle tissues where it plays an important role in maintaining the structural integrity of muscle fibers. Its interaction with other proteins at the Z-disc is important for muscle function.
Biological function summary

Myozenin 2 contributes to the regulation of muscle contraction and signal transduction within muscle cells. It forms part of a multiprotein complex at the Z-disc where it interfaces with proteins such as telethonin and myotilin. This positioning aids in the transmission of mechanical and signaling information necessary for muscle adaptation and maintenance.

Pathways

Myozenin 2 has roles in the regulation of muscle hypertrophy and myopathy pathways. This protein interacts closely with calcineurin a phosphatase involved in calcium-dependent signaling pathways that regulate muscle growth and differentiation. Additionally Myozenin 2 can affect protein kinase C pathways which are essential for modulating the cellular responses to mechanical stress in muscle tissues.

Myozenin 2 is linked to conditions such as familial hypertrophic cardiomyopathy and muscular dystrophy. Changes in the expression or function of Myozenin 2 can lead to disrupted Z-disc integrity and impaired muscle function. In cardiomyopathy Myozenin 2's interaction with calcineurin becomes relevant as alterations in this interaction may contribute to the pathogenesis. In muscular dystrophy its relationship with telethonin becomes significant influencing the structural stability of muscle cells.
