Immunocytochemistry/ Immunofluorescence - Anti-Myozenin 2 antibody (AB237053)
HeLa (Human epithelial cell line from cervix adenocarcinoma) cells labeling Myozenin 2 (green) using ab237053 at 1/100 dilution in ICC/IF, followed by an Alexa-Fluor®488-conjugated goat anti-rabbit IgG (H+L). Countersatined with DAPI (blue).
The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.

Paraffin-embedded human heart tissue stained for Myozenin 2 using ab237053 at 1/300 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Myozenin 2 contributes to the regulation of muscle contraction and signal transduction within muscle cells. It forms part of a multiprotein complex at the Z-disc where it interfaces with proteins such as telethonin and myotilin. This positioning aids in the transmission of mechanical and signaling information necessary for muscle adaptation and maintenance.
Pathways
Myozenin 2 has roles in the regulation of muscle hypertrophy and myopathy pathways. This protein interacts closely with calcineurin a phosphatase involved in calcium-dependent signaling pathways that regulate muscle growth and differentiation. Additionally Myozenin 2 can affect protein kinase C pathways which are essential for modulating the cellular responses to mechanical stress in muscle tissues.
