Natural Killer (NK) Cell Activation Structures are essential components responsible for triggering the killing activity of NK cells. These structures are primarily located on the surface of NK cells where they interact with ligands on target cells. The mass of these activation structures can vary as they often comprise clusters of proteins such as CD16 also known as Fc receptor IIIA and their associated molecules. CD16 has an estimated molecular weight of around 50 kDa. NK cell activation structures are expressed across various tissues where immune surveillance is necessary.
Biological function summary

These structures serve to initiate the cytotoxic response of NK cells by recognizing stressed cells and those lacking "self" markers. When NK cell activation structures engage target cells they form part of a complex that includes proteins such as NKG2D and NKp46. These complexes recognize specific ligands on potential target cells facilitating the release of cytotoxic granules and cytokines. This biological function ensures that NK cells can target and eliminate infected or cancerous cells while sparing normal cells.

Pathways

NK cell activation structures play an important role in the immune response pathways specifically the natural cytotoxicity pathway. They interact with signaling molecules like DAP10 and DAP12 which transmit activation signals to the interior of the NK cells. This signal transduction enhances the ability of NK cells to respond to infections and cancerous changes. Another pathway is the Fc receptor pathway in which CD16 is associated with the activation of NK cells through antibody-dependent cellular cytotoxicity (ADCC).

Activating structures of NK cells have connections to conditions such as cancer and autoimmune diseases. In cancer defective NK cell activation can lead to insufficient immune surveillance allowing tumor cells to proliferate. Malfunctions in these structures potentially linked with proteins like MHC class I chain-related proteins can also result in an autoimmune response where NK cells mistakenly target healthy tissue. Understanding and targeting these structures might lead to improved therapeutic strategies for modulating immune responses in these diseases.
