Biological function summary
These structures serve to initiate the cytotoxic response of NK cells by recognizing stressed cells and those lacking "self" markers. When NK cell activation structures engage target cells they form part of a complex that includes proteins such as NKG2D and NKp46. These complexes recognize specific ligands on potential target cells facilitating the release of cytotoxic granules and cytokines. This biological function ensures that NK cells can target and eliminate infected or cancerous cells while sparing normal cells.
Pathways
NK cell activation structures play an important role in the immune response pathways specifically the natural cytotoxicity pathway. They interact with signaling molecules like DAP10 and DAP12 which transmit activation signals to the interior of the NK cells. This signal transduction enhances the ability of NK cells to respond to infections and cancerous changes. Another pathway is the Fc receptor pathway in which CD16 is associated with the activation of NK cells through antibody-dependent cellular cytotoxicity (ADCC).
