NLRP13 also known as NOD-like receptor family pyrin domain containing 13 is a protein involved in the regulation of inflammatory processes. It has a molecular weight of approximately 120 kDa. NLRP13 is expressed in various tissues with notable expression in immune cells such as macrophages and dendritic cells. Mechanistically NLRP13 belongs to the NOD-like receptor (NLR) family and plays an important role in sensing intracellular pathogens and cellular stress signals.
Biological function summary

NOD-like receptors such as NLRP13 participate in the formation of inflammasomes which are multiprotein complexes that activate inflammatory responses. NLRP13 functions as a sensor that detects specific signals and associates with other proteins to assemble the inflammasome. This assembly leads to the activation of caspases particularly caspase-1 which then cleaves pro-inflammatory cytokines like IL-1β resulting in their active forms. These cytokines are important for mounting effective immune responses against infections.

Pathways

NLRP13 plays a role in the innate immune signaling pathways specifically in the inflammasome pathway. This pathway is essential for detecting pathogens and initiating early immune responses. NLRP13 interacts with other NOD-like receptors and inflammasome components such as ASC (Apoptosis-associated speck-like protein containing a CARD) to propagate these immune signals. This orchestration leads to the appropriate inflammatory response needed to control infections.

Altered NLRP13 function associates with inflammatory and autoimmune conditions. Dysregulation may contribute to gout where excessive inflammatory responses cause joint pain and damage.
