추가 정보

OGDHL also known as Oxoglutarate Dehydrogenase-Like protein plays a significant role in the mitochondrial matrix where it is a part of the alpha-ketoglutarate dehydrogenase complex. This enzyme with a molecular mass of approximately 116 kDa catalyzes the conversion of alpha-ketoglutarate into succinyl-CoA and carbon dioxide. It predominantly expresses in tissues with high metabolic activity such as the brain heart and skeletal muscle where efficient energy production is critical.
Biological function summary

OGDHL facilitates the regulation of the tricarboxylic acid (TCA) cycle an essential component of cellular respiration. It forms a part of a multi-enzyme complex that ensures the continuation of the TCA cycle by contributing to the oxidation of carbohydrates and the generation of energy. The activity of OGDHL directly affects the flux of the cycle impacting cellular metabolic rates and bioenergetics significantly. The integrity and functionality of this complex are important for maintaining energy homeostasis within cells.

Pathways

OGDHL is deeply entwined with the metabolic processes of the TCA cycle and the electron transport chain. Through the TCA cycle it associates with essential respiratory enzymes contributing to ATP production. Moreover its function closely aligns with proteins such as NADH-dehydrogenase which plays a critical role in facilitating electron transfer and oxidative phosphorylation. This connection is vital for sustaining cellular energy needs and for adapting to changing metabolic demands.

Dysregulation of OGDHL activity links to metabolic disorders and neurodegenerative diseases. Deficiencies or malfunctions in OGDHL have associations with conditions such as Leigh syndrome a severe neurological disorder and Alzheimer's disease where energy metabolism is disrupted. In these cases interactions with proteins like amyloid precursor protein and tau become significant as they are often involved in the pathophysiology of such conditions further emphasising OGDHL's importance in maintaining neuronal health and metabolic stability.

타겟 정보

2-oxoglutarate dehydrogenase (E1-like) component of the 2-oxoglutarate dehydrogenase multienzyme complex (OGDHC) which mediates the decarboxylation of alpha-ketoglutarate in the tricarboxylic acid cycle. The OGDHC complex catalyzes the overall conversion of 2-oxoglutarate to succinyl-CoA and CO(2) while reducing NAD(+) to NADH (By similarity). The OGDHC complex is mainly active in the mitochondrion (By similarity). Involved in the inhibition of cell proliferation and in apoptosis (PubMed : 23152800, PubMed : 31175094).
