OXSR1 also known as oxidative stress response 1 or OSR1 is a serine/threonine kinase with a mass of approximately 58 kDa. It plays a mechanical role in regulating cellular responses to environmental stress especially oxidative stress. OXSR1 is expressed in several tissues including the heart kidney and brain indicating its importance across different physiological contexts. The protein functions by activating downstream substrates involved in cellular protection and homeostasis.
Biological function summary

Oxidative stress response kinase 1 participates in cellular defense mechanisms against oxidative damage. OXSR1 does not form part of a larger complex but has unique roles in signaling pathways helping to defend cells from oxidative stress. This protein kinase modulates ion channels and transporters participating in processes that stabilize the cellular environment and maintain ionic balance important for cell survival under stress conditions.

Pathways

OXSR1 plays an important role in the oxidative stress response and the MAPK signaling pathway. The protein interacts with and is regulated by STE20 family kinases including SPAK which are partners in regulating cellular stress responses. These pathways involve phosphorylation events leading to changes in cellular activity therefore supporting adaption to stressful conditions. OXSR1 acts as a mediator in connecting upstream signals to downstream cellular effects.

Oxidative stress response kinase 1 is associated with hypertension and neurodegenerative diseases. Variations or dysfunction in OXSR1 can disrupt ion homeostasis and cellular stress response contributing to the pathogenesis of these conditions. The kinase's interaction with SPAK is significant in these disorders where altered activity influences blood pressure regulation and neuronal survival. Studies continue investigating its potential role in therapeutic interventions for these disorders.

Specifically recognizes and binds proteins with a RFXV motif (PubMed : 16669787, PubMed : 17721439, PubMed : 21321328). Acts downstream of WNK kinases (WNK1, WNK2, WNK3 or WNK4) : following activation by WNK kinases, catalyzes phosphorylation of ion cotransporters, such as SLC12A1/NKCC2, SLC12A2/NKCC1, SLC12A3/NCC, SLC12A5/KCC2 or SLC12A6/KCC3, regulating their activity (PubMed : 17721439). Mediates regulatory volume increase in response to hyperosmotic stress by catalyzing phosphorylation of ion cotransporters SLC12A1/NKCC2, SLC12A2/NKCC1 and SLC12A6/KCC3 downstream of WNK1 and WNK3 kinases (PubMed : 16669787, PubMed : 21321328).
