CD45RB also known as protein tyrosine phosphatase receptor type C (PTPRC) is a membrane-associated protein found prominently on all nucleated hematopoietic cell surfaces. This protein has a molecular mass that ranges between 180-220 kDa depending on the variable splicing. Importantly CD45RB is a component of the CD45 group playing an important role in regulating antigen receptor signaling in lymphocytes affecting both T-cell and B-cell function. Its expression occurs mainly in hematopoietic tissues such as the bone marrow lymph nodes and spleen.

Biological function summary

CD45RB functions as an integral transmembrane protein-tyrosine phosphatase and does not operate as part of a complex but rather acts directly on its substrates. It regulates the phosphorylation state of Src family kinases that are necessary for antigen receptor signaling. Protein tyrosine-protein phosphatase required for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN (By similarity). Dephosphorylates LYN, and thereby modulates LYN activity. Interacts with CLEC10A at antigen presenting cell-T cell contact; CLEC10A on immature dendritic cells recognizes Tn antigen-carrying PTPRC/CD45 receptor on effector T cells and modulates T cell activation threshold to limit autoreactivity.
