PI4KA also known as phosphatidylinositol 4-kinase alpha is an enzyme responsible for phosphorylating phosphatidylinositol to produce phosphatidylinositol 4-phosphate (PI4P). The protein has a mass of approximately 230 kDa. PI4KA is expressed broadly with significant levels in the brain heart and other tissues. It plays an important role as a regulator of cellular lipid metabolism and intracellular signaling.

Biological function summary

PI4KA functions in the context of membrane dynamics and acts as part of a multi-subunit complex. This protein is important in maintaining the proper lipid composition of the Golgi apparatus and other organelles which is essential for vesicle trafficking. By generating PI4P PI4KA serves as an important mediator in the distribution and dynamics of cellular membranes aiding in various cellular processes including endocytosis and autophagy. Its activity impacts several downstream signaling pathways and cellular activities.

Pathways

PI4KA is a significant contributor to the phosphoinositide signaling pathway which plays an essential role in regulating intracellular processes. It interacts with phosphatidylinositol-45-bisphosphate (PIP2) and phospholipase C supporting the synthesis of further derivatives that modulate calcium influx and other cellular responses. Additionally PI4KA is involved in the maintenance and function of the secretory pathway where it coordinates with proteins such as Osh1 and CERT influencing lipid exchange and protein trafficking.
