Immunocytochemistry/ Immunofluorescence - Anti-PIN4 antibody [EPR10033] - BSA and Azide free (AB238927)
Immunocytochemistry/ Immunofluorescence analysis of HepG2 (human hepatocellular carcinoma epithelial cell) cells labeling PIN4 with purified ab155283 at 1/150 dilution (10 μg/mL). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 (2.5 μg/mL). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1/1000 (2 μg/mL) dilution. DAPI (blue) was used as nuclear counterstain. ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 (2.5 μg/mL) was used as the secondary antibody only control.

PIN4 is involved in several critical cellular processes such as cell cycle regulation and RNA processing. It forms part of a complex with other proteins that influence cell cycle progression and check points. Its role in these processes points to its functional versatility in maintaining cellular homeostasis and modulating interactions with other proteins necessary for DNA synthesis and repair.
Pathways
PIN4 interacts with key signaling cascades such as the cell cycle pathway and the ubiquitin proteasome pathway. In the cell cycle pathway it influences progression through the modulation of phosphorylation states of its substrates. In the ubiquitin proteasome pathway PIN4 impacts protein degradation and related proteins include cyclin-dependent kinases (CDKs) and ubiquitin ligases which together regulate cell division and proteostasis.
