The phosphatidylserine decarboxylase (PISD) also called PSD is an enzyme that plays an important role in lipid metabolism within the cell. Its function centers on converting phosphatidylserine to phosphatidylethanolamine an important step in the synthesis of essential phospholipids. PISD with a molecular weight of approximately 45 kDa is expressed in multiple tissues with significant expression found in the mitochondria. This expression highlights its involvement in critical cellular processes related to membrane formation and maintenance.
Biological function summary

PISD acts on phosphatidylserine and contributes to the biosynthesis of phosphatidylethanolamine. The enzyme operates within a mitochondrial lipid biosynthetic pathway often forming part of a protein complex that ensures efficient catalytic activity. By facilitating phospholipid production PISD spans a range of cellular functions including structural support and signaling within the cell membrane. This lipid biosynthetic role is essential for maintaining cellular integrity and function.

Pathways

PISD operates within the glycerophospholipid metabolism and phospholipid biosynthesis pathways. These pathways support essential cellular operations such as membrane fluidity and cell signaling. Within these processes PISD collaborates with proteins like phosphatidylserine synthase 1 (PSS1) which synthesizes phosphatidylserine therefore providing the substrate for PISD activity. Alongside these enzymes the interplay in these pathways ensures the controlled production of phospholipids necessary for cellular homeostasis.

PISD dysfunction associates with metabolic and neurodegenerative conditions. Altered activity of PISD has been linked to mitochondrial disorders where inefficiencies in phospholipid metabolism disrupt mitochondrial function. This disruption can implicate proteins such as cardiolipin synthase which collaborates in the maintenance of mitochondrial integrity. Additionally PISD's role in lipid metabolism can influence conditions like non-alcoholic fatty liver disease where perturbations in lipid homeostasis lead to liver dysfunction. Understanding PISD's involvement offers insights into potential therapeutic targets for these diseases.

타겟 정보

Catalyzes the formation of phosphatidylethanolamine (PtdEtn) from phosphatidylserine (PtdSer) (PubMed : 30488656, PubMed : 30858161). Plays a central role in phospholipid metabolism and in the interorganelle trafficking of phosphatidylserine. May be involved in lipid droplet biogenesis at the endoplasmic reticulum membrane (By similarity).
