PRIM2 was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) using ab241990 at 6 μg per reaction. Western blot was performed from the immunoprecipitate using ab241990 at 0.4 μg/ml.

Lane 1 : ab241990 IP in HEK-293T whole cell lysate.
Lane 2 : Control IgG in HEK-293T whole cell lysate.

Detection : Chemiluminescence with an exposure time of 30 seconds.

The PRIM2 protein also known as DNA primase large subunit plays an essential role in DNA synthesis. With a mass of approximately 58 kDa PRIM2 synthesizes short RNA primers needed for DNA polymerase to start replication. This protein is mainly expressed in tissues with high replication activity including the thymus and bone marrow as well as during the S phase of the cell cycle in various cell types. The PRIM2 protein is critical during the initiation of DNA replication binding to single-stranded DNA to form an RNA-DNA hybrid that DNA polymerases extend.
Biological function summary

DNA primase large subunit forms a complex with the smaller subunit PRIM1 creating the DNA primase enzyme. This complex is part of the larger DNA polymerase alpha-primase complex which is fundamental for the initiation of DNA replication. Primase activity ensures that replication begins accurately and efficiently enabling cells to maintain genomic integrity during cell division. By synthesizing RNA primers PRIM2 facilitates the transition from the initiation to the elongation phase of DNA replication.

Regulatory subunit of the DNA primase complex and component of the DNA polymerase alpha complex (also known as the alpha DNA polymerase-primase complex) which play an essential role in the initiation of DNA synthesis (PubMed : 17893144, PubMed : 25550159, PubMed : 26975377, PubMed : 9705292). During the S phase of the cell cycle, the DNA polymerase alpha complex (composed of a catalytic subunit POLA1, an accessory subunit POLA2 and two primase subunits, the catalytic subunit PRIM1 and the regulatory subunit PRIM2) is recruited to DNA at the replicative forks via direct interactions with MCM10 and WDHD1 (By similarity). The primase subunit of the polymerase alpha complex initiates DNA synthesis by oligomerising short RNA primers on both leading and lagging strands (PubMed : 17893144). These primers are initially extended by the polymerase alpha catalytic subunit and subsequently transferred to polymerase delta and polymerase epsilon for processive synthesis on the lagging and leading strand, respectively (By similarity). In the primase complex, both subunits are necessary for the initial di-nucleotide formation, but the extension of the primer depends only on the catalytic subunit (PubMed : 17893144, PubMed : 25550159). Binds RNA : DNA duplex and coordinates the catalytic activities of PRIM1 and POLA2 during primase-to-polymerase switch.
