Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Prostaglandin D Synthase (Lipocalin)/PDS antibody [EP12357] - BSA and Azide free (AB236119)
The human glioma paraffin-embedded sections were Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20 mins and labeling ab182141 at 1/200 dilution for 30 mins at room temperature, then followed by Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with hematoxylin.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab182141).

Immunocytochemistry/ Immunofluorescence - Anti-Prostaglandin D Synthase (Lipocalin)/PDS antibody [EP12357] - BSA and Azide free (AB236119)
HepG2 cells were fixed in 4% PFA and permeabilized with 0.1% Triton X-100. Primary antibody, ab182141 at 1/200 was incubated overnight at 4° C, followed by AlexaFluor® 488-conjugated Goat anti- Rabbit secondary antibody (ab150081) at 1/1000 dilution at RT for 45 min. ab195889 AlexaFluor® 594-conjugated Anti-alpha Tubulin, used as a counterstain at 1/200 dilution, was co-incubated with ab182141 overnight at 4° C. Nucleus were visualized using DAPI.

Perform heat mediated antigen retrieval with EDTA buffer pH 9 before commencing with IHC staining protocol.

Blocking and diluting buffer and concentration : 5% NFDM/TBST.
In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) staining at 1/1000,000 dilution.
This antibody has been reported to work in western blot with cell lysates by some customers. However, this result could not be reproduced in our latest internal testing. Human cerebellum is recommended as a positive control.

Biological function summary
This protein PDS participates in various physiological processes including inflammation and sleep regulation. It operates independently and is not typically part of larger protein complexes. PGDS contributes to the homeostasis of biological membranes and acts as a neuromodulator in the brain. Its function extends to mediating the actions of the prostaglandin D2 it produces affecting processes like allergic reactions and maintaining normal reproductive physiological functions.
