PTPN18 also known as protein tyrosine phosphatase non-receptor type 18 or BDP1 functions as a tyrosine phosphatase enzyme involved in signaling pathways by removing phosphate groups from tyrosine residues on proteins. This protein has a molecular mass of approximately 48 kDa. It is expressed in various tissues with particular presence in the brain liver and skeletal muscle. By modulating phosphorylation states PTPN18 plays a vital role in cellular signal transduction.
Biological function summary

PTPN18 influences cellular processes such as growth differentiation and apoptosis by regulating phosphorylation events. This enzyme acts in concert with other signaling molecules although it does not form specific complexes itself. PTPN18 interacts with substrates like the HER2/ERBB2 receptor impacting critical pathways regulating cell proliferation and survival. These interactions allow PTPN18 to exert control over cellular responses helping maintain homeostasis within biological systems.

Pathways

PTPN18 is an important modulator within the ERK/MAPK and PI3K/Akt signaling cascades. Within the ERK/MAPK pathway PTPN18 interacts with proteins such as SHC1 and GRB2 to help regulate cellular activities like division and differentiation. In the PI3K/Akt pathway PTPN18 affects cell survival and metabolism by regulating components in the signaling cascade. These interactions establish PTPN18 as a critical player in transmitting extracellular signals to the intracellular machinery.

PTPN18 has connections to cancer and metabolic disorders. It is implicated in the progression and prognosis of certain cancers including breast cancer where it modulates the activity of the ERBB2 receptor. By influencing phosphorylation PTPN18 can impact tumor growth and response to treatment. Additionally alterations in PTPN18 activity may contribute to metabolic diseases through its effects on receptor signaling and cellular metabolism. Understanding these interactions highlights PTPN18's relevance in disease mechanisms and the potential for therapeutic interventions.
