PZP also known as Pregnancy Zone Protein is a critical glycoprotein with a molecular weight of approximately 360 kDa. It is mainly expressed in the liver but also found in the blood plasma particularly during pregnancy. PZP is similar in structure to alpha-2-macroglobulin and functions by inhibiting proteases. This protein binds and inactivates various proteinases showing its role in modulating enzymatic activity in the bloodstream.
Biological function summary

PZP contributes to immune response modulation during pregnancy by forming complexes with proteinases. These complexes prevent the degradation of other proteins helping to protect the fetus and maintain maternal immune tolerance. PZP exists as a tetramer in its active form allowing it to effectively trap and neutralize proteinases. Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase.
