Rad22 also known as RPA1 or replication protein A1 is a protein involved in DNA repair processes. It has a molecular mass of approximately 29 kDa. Rad22 is expressed in eukaryotic cells and has a critical role in maintaining the integrity of genetic material. The protein interacts with single-stranded DNA (ssDNA) during repair and replication processes forming a nucleoprotein complex that stabilizes ssDNA intermediates and protect them from nuclease degradation.
Biological function summary

Rad22 contributes to homologous recombination an essential mechanism for repairing double-strand breaks in DNA. Rad22 is a part of a larger multiprotein complex that includes Rad51 and other early recombination factors. This complex plays a role in the search for homology and the strand pairing stages of homologous recombination. Rad22's interaction with Rad51 ensures accurate repair by facilitating the invasion of the broken DNA strand into the homologous sequence.

Pathways

Cellular processes involving Rad22 engage in the homologous recombination repair pathway and DNA replication stress response. Rad22 acts synergistically with Rad51 and other recombination mediators to initiate and propagate homologous recombination. These processes ensure the correct repair of DNA to prevent genomic instability which can lead to cell cycle arrest or apoptosis.

Rad22's function impacts conditions related to genomic instability such as cancer and Bloom syndrome. Disruptions in Rad22 activity can lead to inefficient DNA repair contributing to the accumulation of mutations that drive tumorigenesis. Furthermore Rad22 interacts with other proteins like BRCA2 in tumor suppression pathways. In Bloom syndrome a disorder characterized by high rates of chromosome breakage Rad22 plays a vital part in DNA repair and any malfunctions in its activity connect to the disease phenotype.
