Paraffin-embedded human liver tissue stained for RECK using ab238162 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

HepG2 (human liver hepatocellular carcinoma cell line) cells stained for RECK (green) using ab238162 at 1/100 dilution in ICC/IF, followed by Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Counterstained with DAPI.

RECK also known as Reversion-inducing-cysteine-rich protein with kazal motifs is a 110 kDa membrane-anchored glycoprotein. It plays a role in inhibiting matrix metalloproteinases (MMPs) like MMP-2 and MMP-9 which are involved in extracellular matrix remodeling. RECK is widely expressed in various tissues including the brain heart and muscles. It interacts with other membrane proteins to modulate different cellular functions playing a part in maintaining the structure of tissues.

Functions together with ADGRA2 to enable brain endothelial cells to selectively respond to Wnt7 signals (WNT7A or WNT7B) (PubMed : 28289266, PubMed : 30026314). Plays a key role in Wnt7-specific responses : required for central nervous system (CNS) angiogenesis and blood-brain barrier regulation (By similarity).
