RLTPR also known as CARMIL2 is a protein involved in T-cell receptor (TCR) signaling. It has a molecular mass of approximately 150 kDa. It is expressed mainly in lymphoid tissues including the thymus and spleen playing an important role in the immune response. RLTPR serves as an adapter protein mediating interactions that are important for signal transduction in immune cells.
Biological function summary

RLTPR supports the activation and proliferation of T-cells by interacting with other intracellular molecules. It forms a complex with the scaffold protein CARD11 and the coiled-coil domain of caspase recruitment domains aiding in the assembly of the immune synapse. This assembly promotes effective T-cell receptor signaling facilitating the immune system's capacity to recognize and react to foreign antigens.

Pathways

RLTPR plays a significant role in the NF-kB signaling pathway which regulates the expression of genes involved in the immune and inflammatory responses. It interacts with proteins such as BCL10 and MALT1 within this pathway contributing to the activation of transcription factors that drive gene expression. RLTPR is also linked to the mTORC1 pathway correlating with processes of cell growth and metabolism.

Disruptions of RLTPR function have been implicated in immunodeficiency and autoimmune disorders. Deficiencies in RLTPR can lead to compromised immune systems often resulting in conditions such as common variable immunodeficiency (CVID). In the context of autoimmune disorders RLTPR abnormalities may contribute to the misregulation of T-cell activation linking it to related proteins such as RHOA that are involved in cell signaling and immune responses.
