SARDH also known as sarcosine dehydrogenase is an enzyme involved in the metabolism of sarcosine a derivative of the amino acid glycine. SARDH has a molecular mass of approximately 98 kDa. This enzyme catalyzes the oxidative demethylation of sarcosine to produce glycine providing an important step in the catabolic pathway of one-carbon units. SARDH is mainly expressed in the mitochondria of the liver and kidney highlighting its role in these organs' metabolism.
Biological function summary

Sarcosine dehydrogenase is responsible for maintaining the balance of sarcosine and related metabolites. By converting sarcosine to glycine SARDH plays a role in the methionine cycle and folate-mediated one-carbon metabolism. This enzyme does not function as part of a larger protein complex but interacts independently within cellular metabolism processes. Proper functioning of SARDH helps in the regulation of cellular methylation reactions as it is interconnected with other metabolic pathways that manage methyl groups.

Pathways

SARDH is an important component of the glycine and serine metabolism pathway. This pathway is integral to the one-carbon metabolic network which is important for nucleic acid synthesis and cell division. SARDH is related to other enzymes like dimethylglycine dehydrogenase (DMGDH) through the catabolic processes of different methylated amino acids. Additionally it participates in the methionine cycle closely linked with enzymes such as methionine synthase emphasizing its interconnectedness in maintaining amino acid homeostasis.

Mutations or deficiencies in SARDH activity have been linked to nonketotic hyperglycinemia a rare metabolic disorder characterized by elevated levels of glycine in body fluids. This disorder can lead to neurological complications due to glycine's role as a neurotransmitter. Another condition associated with SARDH is sarcosinemia where sarcosine accumulates abnormally due to improper sarcosine degradation. SARDH's dysfunction may also be related to prostate cancer through the interaction with enzymes in the methylation cycle indicating its potential role in tumorigenesis.
