Sly1 also known as suppressor of Ypt1 homolog 1 functions as a protein involved in vesicle fusion. It has a molecular mass of approximately 67 kDa and serves as an important element in vesicular trafficking processes. Expressed in various tissues Sly1 primarily resides in the cytoplasm and the Golgi apparatus. It facilitates the fusion of transport vesicles to compartment membranes by binding to SNARE proteins therefore playing a role in maintaining normal cellular transport operations.
Biological function summary

Sly1 influences cellular transport by being part of the SM (Sec1/Munc18) protein family. It regulates SNARE complex assembly ensuring accurate vesicle docking and fusion events. Sly1 interacts with different t-SNAREs such as Syntaxin 5 modulating their functions and energy coupling during vesicle fusion. Proper action of Sly1 is necessary for maintaining cellular organization and intracellular communication influencing the efficiency of endocytotic and exocytotic pathways.

Pathways

Sly1 participates actively within the vesicle transport mechanisms linked by SNARE proteins. It plays a fundamental role in the secretory pathway affecting protein and lipid trafficking. Impairments in Sly1 expression or function relate to neurodegenerative diseases such as Alzheimer's disease. These conditions associate with defects in vesicular trafficking pathways as evidenced by altered Sly1 interactions with proteins like Syntaxin 5. Such interruptions can cause misprocessing and accumulation of proteins within neurons. Another disorder linked to Sly1 is developmental brain disorder where disruptions in vesicular fusion contribute to improper neuronal connections.
