Biological function summary
SPEN plays a role in transcriptional repression and gene silencing by being part of a multiprotein complex. It interacts with nuclear receptors and RNA polymerase II inhibiting the transcriptional activity of target genes. SPEN helps regulate critical cellular processes such as differentiation and proliferation. It acts as a transcriptional corepressor by interacting with nucleic acids and influencing chromatin structure impacting gene expression in a context-dependent manner.
Pathways
SPEN holds an important role in the Notch signaling pathway. This pathway is important for cell fate determination and tissue homeostasis. SPEN also interacts with the estrogen receptor signaling pathway where it can influence the transcriptional outcome. Through these pathways SPEN associates with proteins such as RBPJ a transcriptional regulator in the Notch pathway and ERα (estrogen receptor alpha) a vital component in the estrogen signaling pathway.
