SPEN also known as Split Ends or SHARP is a large nuclear protein with a mass of approximately 400 kDa. It is expressed in various tissues including the brain heart and lungs. Mechanically SPEN functions as an RNA-binding protein with C-terminal SPOC (Spen Paralog and Ortholog C-terminal) domains which allow it to interact with other cellular components. These interactions regulate transcription by modulating the activity of coactivators and corepressors at specific gene sites.
Biological function summary

SPEN plays a role in transcriptional repression and gene silencing by being part of a multiprotein complex. It interacts with nuclear receptors and RNA polymerase II inhibiting the transcriptional activity of target genes. SPEN helps regulate critical cellular processes such as differentiation and proliferation. It acts as a transcriptional corepressor by interacting with nucleic acids and influencing chromatin structure impacting gene expression in a context-dependent manner.

Pathways

SPEN holds an important role in the Notch signaling pathway. This pathway is important for cell fate determination and tissue homeostasis. SPEN also interacts with the estrogen receptor signaling pathway where it can influence the transcriptional outcome. Through these pathways SPEN associates with proteins such as RBPJ a transcriptional regulator in the Notch pathway and ERα (estrogen receptor alpha) a vital component in the estrogen signaling pathway.

SPEN has connections to certain cancers particularly breast cancer and neurological conditions like autism spectrum disorders. In breast cancer SPEN can interact with ERα influencing the proliferation of cancer cells through estrogen receptor signaling. In neurological conditions dysregulation of SPEN's interaction with proteins involved in neural development might contribute to disease mechanisms. In osteoblasts, supports transcription activation : synergizes with RUNX2 to enhance FGFR2-mediated activation of the osteocalcin FGF-responsive element (OCFRE) (By similarity).
