Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-ST3GAL4/STZ antibody (AB238742)

Formalin-fixed, paraffin-embedded human cervical cancer tissue stained for ST3GAL4/STZ using ab238742 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

ST3GAL4 also known as STZ is a glycosyltransferase enzyme responsible for the addition of sialic acid to galactose-containing substrates specifically at the third carbon position of the galactose unit. This enzyme has a molecular mass of approximately 43 kDa. It is expressed mainly in the human liver pancreas and brain. By catalyzing the transfer of sialic acid ST3GAL4 influences the charge and recognition potential of glycoproteins and glycolipids on the cell surface.
Biological function summary

ST3GAL4 contributes to the synthesis of sialylated glycoconjugates which plays a significant role in cell-cell interactions signaling and immune response modulation. It is involved in the creation of the sialyl Lewis X (sLe^x) epitope an important component in selectin-mediated adhesion processes. ST3GAL4 does not function as part of a larger enzymatic complex but orchestrates critical modification of glycoconjugates on its own. Its activity affects how cells communicate and interact within the physiological environment.

Pathways

ST3GAL4 operates within the glycosylation pathway specifically influencing the N-glycan biosynthesis pathway. Its action is important for the formation of complex glycan structures on proteins and lipids influencing their stability and function. ST3GAL4 is related to other sialyltransferases such as ST3GAL1 which also contribute to the diversity of glycan structures in cells. The proper function of these pathways ensures the correct formation of glycan codes critical for cellular communication.

Aberrant activity or expression of ST3GAL4 associates with cancer and inflammatory diseases. Changes in the sialylation pattern mediated by ST3GAL4 alter cell adhesion and migration contributing to cancer metastasis and progression. In inflammatory disorders altered glycosylation can lead to abnormalities in immune cell interactions exacerbating the disease state. ST3GAL4 connects with proteins like selectin affecting cell adhesion dynamics in both oncological and inflammatory contexts.

타겟 정보

A beta-galactoside alpha2-3 sialyltransferase involved in terminal sialylation of glycoproteins and glycolipids (PubMed : 8288606, PubMed : 8611500, PubMed : 37632720). Catalyzes the transfer of sialic acid (N-acetyl-neuraminic acid; Neu5Ac) from the nucleotide sugar donor CMP-Neu5Ac onto acceptor Galbeta-(1->3)-GalNAc- and Galbeta-(1->4)-GlcNAc-terminated glycoconjugates through an alpha2-3 linkage (PubMed : 8288606, PubMed : 8611500). Plays a major role in hemostasis. Responsible for sialylation of plasma VWF/von Willebrand factor, preventing its recognition by asialoglycoprotein receptors (ASGPR) and subsequent clearance. Regulates ASGPR-mediated clearance of platelets (By similarity). Participates in the biosynthesis of the sialyl Lewis X epitopes, both on O- and N-glycans, which are recognized by SELE/E-selectin, SELP/P-selectin and SELL/L-selectin. Essential for selectin-mediated rolling and adhesion of leukocytes during extravasation (PubMed : 25498912). Contributes to adhesion and transendothelial migration of neutrophils likely through terminal sialylation of CXCR2 (By similarity). In glycosphingolipid biosynthesis, sialylates GM1 and GA1 gangliosides to form GD1a and GM1b, respectively (PubMed : 8288606). Metabolizes brain c-series ganglioside GT1c forming GQ1c (By similarity). Synthesizes ganglioside LM1 (IV3Neu5Ac-nLc4Cer), a major structural component of peripheral nerve myelin (PubMed : 8611500).
