ab50595 staining TGN46 in HepG2 cells (top panel - positive control) and HeLa cells (bottom panel - negative control, low TGN46 expression). The cells were fixed with 4% paraformaldehyde (10 min) then permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated with ab50595 at 1μg/ml concentration and ab7291 (Mouse monoclonal to alpha Tubulin) at 1/1000 dilution overnight at 4°C followed by a further incubation at room temperature for 1h with a goat secondary antibody to rabbit IgG (Alexa Fluor® 488) (ab150081) at 2 μg/ml (shown in green) and a goat secondary antibody to mouse IgG (Alexa Fluor® 594) (ab150120) at 2 μg/ml (shown in red). Nuclear DNA was labelled in blue with DAPI.

ab50595 staining TGN46 in human HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% paraformaldehyde in PBS for 10 minutes, permeabilized with 0.1% Triton X100 and blocked with 2% BSA for 30 minutes at 25°C. Samples were incubated with primary antibody (1/200 in 2% BSA + 0.1% Triton X100) for 2 hours at 25°C. An Alexa Fluor® 594-conjugated Goat polyclonal to mouse IgG (dilution 1/1500) was used as secondary antibody.

Cells were fixed with formaldehyde followed by blocking in 1% Donkey serum in PBST for 1 hour at 22°C. A donkey-anti-Rb Alexa Fluor® 594 was used as the secondary antibody at 1/200 dilution. TGN46 also known as trans-Golgi network protein 46 is an important membrane protein associated with the trans-Golgi network. The protein has a mass of approximately 46 kDa and plays a role in the sorting and trafficking of proteins to their proper destinations within the cell. TGN46 is widely expressed in various cell types and is commonly used as a Golgi marker in cell biology studies helping to identify Golgi apparatus in cells.
Biological function summary

The protein TGN46 functions in the sorting and transport mechanisms within the cell. It does not function as a part of a large macromolecular complex but it interacts with several other proteins to facilitate the movement of proteins from the Golgi to other sites within the cell. The protein also associates with trafficking components to ensure the correct delivery of proteins to the plasma membrane or lysosomes.

Pathways

The role of TGN46 involves participation in intracellular transport and protein trafficking pathways.
