Immunocytochemistry/ Immunofluorescence - Anti-Thyroid Hormone Receptor beta antibody (AB53170)

ICC/IF image of ab53170 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab53170, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

The Thyroid Hormone Receptor beta (THRB) also known as c-erbA-2 and NR1A2 is a nuclear hormone receptor and transcription factor weighing approximately 55 kDa. THRB binds thyroid hormones and plays a central role in moderating gene expression. Expression of this receptor occurs in various tissues with notable presence in the liver brain and heart. As a transcription factor it influences the transcription of specific target genes upon binding to thyroid hormones primarily thyroxine (T4) and triiodothyronine (T3).
Biological function summary

This receptor regulates critical metabolic processes and plays a role in development. THRB functions as part of a larger receptor complex partnering with retinoid X receptor (RXR) to form a heterodimer. This dimer binds to thyroid hormone response elements (TREs) on DNA modulating the expression of genes involved in metabolism thermogenesis and development of the central nervous system.
