2% paraformaldehyde-fixed U-2 OS (Human bone osteosarcoma epithelial cell line) cells labeling TIPIN with ab236451 (green) at 1/600 dilution in ICC/IF. Cells were treated with siRNA against TIPIN (right panel) or control (left panel). Counter stained with DAPI (blue). Incubated with primary antibody overnight at +4°C.

TIPIN was immunoprecipitated from U-2 OS (Human bone osteosarcoma epithelial cell line) whole cell lysate using 3 μl of ab236451.

Lane 1 : U-2 OS whole cell lysate ~400 μg (Input).

Lane 2 : Control IgG in U-2 OS whole cell lysate.

Lane 3 : ab236451 in U-2 OS whole cell lysate.

For western blotting ab236451 was used at a 1/2000 dilution.

The TIPIN protein also known as Timekeeper Homolog is a 29 kDa protein involved in the regulation of DNA replication and genome stability. It interacts with other proteins to form a complex that plays a role in stabilizing the replication machinery. TIPIN expresses in various tissues including those with high proliferation rates. Researchers study its mechanical role because of its involvement in maintaining the proper functioning of the replication fork a critical part of DNA synthesis.
Biological function summary
