Immunofluorescent analysis of Jurkat cells (4% Paraformaldehyde-fixed, 0.1% tritonX-100 permeabilized) labeling TLT-1 with ab192619 at 1/50 dilution (1.6 μg/ml). A Goat anti rabbit IgG (Alexa Fluor488) at 1/400 dilution (ab150077) was used as secondary antibody. Counterstain : DAPI.

Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

TLT-1 or TREM-like transcript 1 is a receptor expressed on platelets and megakaryocytes. Known for a mass of approximately 27 kDa TLT-1 belongs to the immunoglobulin superfamily. This receptor is a type I transmembrane protein and its expression is found mainly in the alpha-granules of resting platelets. During platelet activation TLT-1 translocates to the cell surface where it can participate in various cellular interactions.
Biological function summary

TLT-1 plays a significant role in hemostasis and inflammation. When considering its biological activity TLT-1 is not part of a larger protein complex but it can bind fibrinogen which facilitates clot formation and stability. This binding assists in the regulation of platelet aggregation and assists modulating the inflammatory response during vascular injury. Through its interactions at the molecular level TLT-1 enhances the overall efficiency of the clotting process.

Pathways

TLT-1 contributes to the hemostatic and pro-inflammatory pathways. It plays a part in the regulation of clot formation by interacting with other platelet surface proteins and fibrinogen. TLT-1 is associated with pathways such as the platelet activation pathway where it cooperates with proteins like P-selectin to modulate responses to vascular injury. Such interactions ensure that TLT-1 functions as an important component in maintaining circulatory system integrity.

TLT-1 connects to conditions such as thrombosis and sepsis. These disorders involve the dysfunction of the hemostatic and immune systems respectively. In thrombosis TLT-1 may contribute to excessive clot formation often interacting with proteins like fibrinogen in the process. During sepsis TLT-1 takes part in the inflammatory response which can exacerbate the clinical symptoms of the condition. Understanding the role of TLT-1 could aid in developing therapeutic targets aimed at modulating both coagulation and inflammation processes.
