Trichohyalin (THH) acts mechanically as a structural protein that plays an important role in hair and skin cell cornification. Expressed in the inner root sheath of hair follicles and in the granular layer of the epidermis it contributes to cell envelope formation. This protein with a molecular mass of approximately 250 kDa is also known as trichohyalin-1. THH is rich in glutamine and lysine which allows it to bind to other proteins and form a stable matrix structure in hair and skin.
Biological function summary

Trichohyalin contributes to the development and maintenance of hair follicle integrity and skin barrier function. As part of the keratinocyte differentiation process it interacts with various keratins and contributes to cell envelope formation by reinforcing cell membranes through cross-linking. The protein forms a part of the cornified cell envelope complex where it binds with filaggrin and loricrin aiding in cellular structural support and resistance to mechanical stress.

Pathways

Trichohyalin integrates into the cellular pathways involved in keratinocyte differentiation and cornification. It participates closely in the epidermal differentiation complex a critical pathway for skin barrier function. Trichohyalin works alongside proteins such as involucrin and filaggrin to facilitate these processes ensuring the maintenance of skin homeostasis and protective barrier formation.

The altered expression or malfunction of trichohyalin links to disorders like ichthyosis vulgaris and hair shaft abnormalities. Researchers observe that defective trichohyalin also relates to structural skin anomalies associated with the malfunction of proteins like loricrin and involucrin.

Intermediate filament-associated protein that associates in regular arrays with keratin intermediate filaments (KIF) of the inner root sheath cells of the hair follicle and the granular layer of the epidermis. It later becomes cross-linked to KIF by isodipeptide bonds. It may serve as scaffold protein, together with involucrin, in the organization of the cell envelope or even anchor the cell envelope to the KIF network. It may be involved in its own calcium-dependent postsynthetic processing during terminal differentiation.
