TRIM17 (Tripartite Motif Containing 17) also known as RNF93 acts as an E3 ubiquitin-protein ligase. This protein mechanically transfers ubiquitin from E2 ubiquitin-conjugating enzymes to specific substrate proteins which marks them for proteasomal degradation. TRIM17 with a mass of approximately 60 kDa exhibits expression in various tissues including the brain testis and spleen. The TRIM family to which TRIM17 belongs shares a common structure of RING finger B-box and coiled-coil domains which are critical for their ligase activity.
Biological function summary

TRIM17 inhibits neuronal survival by inducing neuronal apoptosis playing a significant role in brain development and homeostasis. Although TRIM17 itself is not a part of a large complex it interacts with factors like MCL1 a pro-survival member of the BCL2 family which influences apoptotic signaling pathways. TRIM17 is important in the regulation of protein degradation that modulates apoptotic cascades ensuring the proper turnover of proteins necessary for cell death.

Pathways

TRIM17 interacts with the ubiquitin-proteasome system to regulate proteostasis and neuronal apoptosis. It participates in the JNK signaling pathway where it modulates cellular stress responses. TRIM17 also shows functional interactions with proteins such as p53 which plays a role in oxidative stress response pathways. These pathways highlight its involvement in maintaining the balance between cell survival and death especially in neuronal tissues.

TRIM17 has links to neurodegenerative diseases such as Parkinson's disease and Huntington's disease. Its regulation of apoptosis and protein homeostasis can influence the progression of these conditions. TRIM17 interaction with proteins like PARK2 a gene associated with Parkinson's disease suggests its potential role in the pathophysiology of neurodegenerative disorders offering a target for therapeutic intervention.

E3 ubiquitin ligase that plays important roles in the regulation of neuronal apoptosis, selective autophagy or cell proliferation (PubMed : 19358823, PubMed : 22023800, PubMed : 27562068). Stimulates the degradation of kinetochore ZW10 interacting protein ZWINT in a proteasome-dependent manner, leading to negative regulation of cell proliferation (PubMed : 22023800). Inhibits autophagic degradation of diverse known targets while contributing to autophagy of midbodies. Decreases TRIM41-mediated degradation of ZSCAN2 thereby stimulating alpha-synuclein/SNCA transcription in neuronal cells (By similarity). Prevents the E3 ubiquitin-ligase activity of TRIM28 and its interaction with anti-apoptotic BCL2A1, blocking TRIM28 from ubiquitinating BCL2A1 (PubMed : 19358823).
