추가 정보

Tropomodulin 1 also known as TMOD1 is a capping protein responsible for regulating actin filament dynamics by binding to the pointed end of actin filaments preventing filament elongation and depolymerization. This interaction involves actin-binding sites and tropomyosin-binding regions providing stability to the filament structure. Tropomodulin 1 has an approximate mass of 40 kDa and is expressed in multiple tissues predominantly found in skeletal and cardiac muscles where it plays an essential role in maintaining the integrity of muscle fibers.
Biological function summary

Tropomodulin 1 controls the length of actin filaments impacting muscle contraction and cell motility. It forms part of a larger complex with tropomyosin assisting in stabilizing thin filaments. By effectively managing actin filament architecture Tropomodulin 1 contributes to structural cellular functions supporting cell morphology and intracellular transport. Its interaction with tropomyosin facilitates the proper functioning of contractile systems in muscle cells.

Pathways

Tropomodulin 1 participates in the sarcomere organization and muscle contraction pathways where it ensures the appropriate arrangement of the actin cytoskeleton. Tropomodulin 1’s regulation of actin-tropomyosin interactions affects proteins such as troponin which plays an important role in the regulation of calcium binding during muscle contractions. These pathways are critical for muscle development and proper neuromuscular functionality.

Mutations or dysregulation of Tropomodulin 1 have been implicated in cardiomyopathies and nemaline myopathy. These conditions often result from mismanagement of actin filament lengths affecting muscle fiber stability. Tropomodulin 1’s pathological role often involves disruptions in its binding with tropomyosin and such imbalances can lead to severe muscular disorders. By affecting the actin cytoskeleton Tropomodulin 1 connections to proteins such as actin and tropomyosin become significant in understanding these muscle-related diseases.

타겟 정보

Blocks the elongation and depolymerization of the actin filaments at the pointed end (PubMed : 38168645). The Tmod/TM complex contributes to the formation of the short actin protofilament, which in turn defines the geometry of the membrane skeleton. May play an important role in regulating the organization of actin filaments by preferentially binding to a specific tropomyosin isoform at its N-terminus.
