VBP-1 also known as Von Hippel-Lindau binding protein 1 or prefoldin subunit 3 is a small protein with a molecular mass of approximately 23 kDa. It is expressed in various tissues including the brain heart and skeletal muscle. Mechanically VBP-1 functions as a molecular chaperone facilitating the proper folding of newly synthesized proteins and preventing the aggregation of misfolded proteins. It interacts with different molecular partners to exert its chaperoning activity.
Biological function summary

VBP-1 plays an important role in protein homeostasis particularly by forming part of the prefoldin complex. This complex works alongside the cytosolic chaperonin complex helping with the proper folding of cytoskeletal proteins such as actin and tubulin. By stabilizing these newly synthesized proteins VBP-1 supports cellular structures and intracellular transport systems reflecting its involvement in maintaining cellular integrity.

Pathways

VBP-1 plays a role in the ubiquitin-proteasome pathway and the cellular stress response pathway. In the ubiquitin-proteasome pathway it partners with proteins like HSP70 to target damaged proteins for degradation. In response to cellular stress VBP-1 helps in protein stabilization to prevent harmful aggregation correlating with HSP90 in maintaining cellular homeostasis during stress conditions.

VBP-1 has connections with neurodegenerative diseases such as Alzheimer's disease due to its role in protein folding and aggregation. Misregulation of VBP-1 can lead to the accumulation of neurotoxic protein aggregates. Additionally VBP-1 has been associated with cancer particularly through its interaction with pVHL (von Hippel-Lindau protein). Dysregulation in VBP-1 activity can result in the disruption of the ubiquitin-proteasome system leading to uncontrolled cell growth and cancer development.
