Immunocytochemistry/ Immunofluorescence - Anti-YY1 Antibody [RP23040257] - Nuclear Loading Control (AB313420)
Immunofluorescent analysis of DU 145 cells fixed with 4% formaldehyde reconstituted in 1X PBS for 10 min at room temperature and permeabilized with 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling endogenous YY1 with ab313420 at 1/100 dilution, followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at 1/2000 dilution. Panel a) shows representative cells that were stained for detection and localization of YY1 protein (green), Panel b) is stained for nuclei (blue) using DAPI. Panel c) represents cytoskeletal F-actin staining using Rhodamine Phalloidin at 1/300 dilution. Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of YY1. Panel e) represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Biological function summary
Yin Yang 1 plays an important role in various cellular processes such as cell proliferation differentiation and apoptosis. It is part of a chromatin remodeling complex where it assists in modifying the architecture of chromatin to control gene activity. Due to its ability to bind to diverse DNA motifs YY1 is involved in embryonic development and cellular differentiation. Additionally it interacts with other transcription factors to coordinate the expression of genes critical for cell cycle progression and survival.

It plays a notable role in the Wnt signaling pathway which is vital for controlling cell growth and differentiation.
