The CTX-M-1 beta-lactamase also known as CTX-M or CTX-M protein is a member of the class A beta-lactamases specifically extended spectrum beta-lactamases (ESBLs). This enzyme has a molecular weight of approximately 29 kDa. CTX-M-1 is primarily expressed in gram-negative bacteria particularly Escherichia coli frequently referred to as E. coli beta lactamase. Its primary function is to hydrolyze beta-lactam antibiotics including penicillins and cephalosporins rendering them inactive.
Biological function summary

The CTX-M-1 enzyme plays an important role in antibiotic resistance. It hydrolyzes the beta-lactam ring structure of antibiotics which allows bacteria to survive in environments with these antimicrobials. CTX-M-1 functions as a single protein unit and does not form part of a larger complex. This resistance mechanism contributes to the increased survival of E. coli and other bacteria in clinical settings where antibiotics are often used.

Pathways

CTX-M-1 beta lactamase impacts the antibiotic resistance pathway. It directly contributes to the breakdown of beta-lactam antibiotics which is an important step in resistance pathways. CTX-M-1 often interacts with other antibiotic resistance proteins like TEM and SHV beta-lactamases which also hydrolyze related compounds. These interactions complicate treatment strategies as the proteins together enhance a bacteria's resistance profile.

CTX-M-1 beta lactamase plays a significant role in urinary tract infections (UTIs) and sepsis caused by drug-resistant E. coli. These infections are challenging to treat due to the presence of CTX-M-1 rendering many frontline antibiotics ineffective. The enzyme's activity relates to other proteins such as Klebsiella pneumoniae carbapenemase (KPC) which shares a similar antibiotic resistance role contributing to multidrug-resistant infections.
