E. coli heat labile toxin A also known as LT-A is one component of the heat-labile enterotoxin produced by enterotoxigenic Escherichia coli (ETEC). LT-A has a molecular mass of approximately 27 kDa. This toxin consists of two subunits: the enzymatically active A subunit and the pentameric B subunit. Primarily LT-A gets expressed in the intestinal lumen where ETEC colonizes. LT-A is responsible for the ADP-ribosylation of the alpha subunit of G proteins leading to sustained activation of adenylate cyclase.

E. coli heat labile toxin A plays a significant role in causing traveler's diarrhea by disrupting water and electrolyte absorption in enterocytes. It is part of the ab5 toxin family where it forms a complex with the B subunits to exert its toxic effects. The disruption occurs when the catalytic A subunit enters the host cell after the B subunit binds to the GM1 gangliosides on the cell membrane. The continuous activation of adenylate cyclase increases intracellular cyclic AMP (cAMP) levels leading to chloride ion and water secretion into the intestinal lumen.

The elevation of cAMP levels driven by E. coli heat labile toxin A activates the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel pathway.
