추가 정보

Trigger factor also known as TF or tig acts as a molecular chaperone and peptidyl-prolyl isomerase. It exhibits a mass of approximately 48 kDa. This protein is highly expressed in prokaryotic cells especially in Escherichia coli. Trigger factor associates with ribosomes facilitating the proper folding of nascent polypeptide chains emerging from the ribosomal exit tunnel. It can alternate between ribosome-bound and free states enabling it to assist in post-translational folding tasks.
Biological function summary

Trigger factor prevents aggregation and misfolding of newly synthesized proteins. It plays a significant role in the initial stages of protein folding. It does not operate as part of a multi-protein complex but can function independently or in coordination with other chaperones like DnaK enhancing the overall protein folding capacity within cells. The presence of trigger factor regulates the equilibrium between folding competent and non-competent polypeptides supporting cellular protein quality control.

Pathways

Trigger factor contributes to protein homeostasis and is integral in the protein quality control pathway. This pathway ensures proper folding and functionality of proteins reducing the occurrence of dysfunctional proteins that could lead to cellular stress. Related proteins include DnaK and GroEL which also participate in the unfolded protein response. These proteins collectively maintain proteostasis by managing misfolded proteins and facilitating proper protein folding.

Trigger factor involvement becomes evident in conditions related to protein misfolding such as neurodegenerative diseases. Although trigger factor is not directly linked to human diseases the concept of protein misfolding is critical in disorders like Alzheimer's disease and Parkinson's disease where proteins such as amyloid-beta and alpha-synuclein play a role. Understanding the mechanisms of trigger factor may contribute to strategies aiming to alleviate protein misfolding in these disorders.
