The EBV gp340/220 Envelope Protein also known as the Epstein-Barr virus major glycoprotein gp350/220 plays an important role in the viral entry mechanism. It is approximately 340 kDa in mass and is expressed on the surface of the Epstein-Barr virus (EBV) particles. This glycoprotein mediates the attachment of virions to host cells by binding to the complement receptor type 2 (CR2/CD21) on B lymphocytes which facilitates the viral entry into the host cell. The expression of gp340/220 is restricted to EBV-infected cells and encapsulates the virus playing an integral role in its infectivity.

This glycoprotein promotes viral entry by enhancing the fusion between the viral envelope and the host cell membrane. It is an important component of the viral entry machinery acting in conjunction with other viral proteins such as gp42 to initiate infection. In infectious mononucleosis this glycoprotein participates in binding to CR2/CD21 facilitating viral entry and causing the characteristic symptoms of the disease.
