AR-V7 also known as Androgen Receptor Splice Variant 7 is a 75 kDa protein. It is a variant of the full-length androgen receptor (AR) but lacks the ligand-binding domain due to alternative splicing. This characteristic allows AR-V7 to remain active even in the absence of androgens making it mechanistically significant. AR-V7 expression occurs mainly in prostate tissues where the AR is fundamental to normal and cancerous growth but AR-V7 is also reported in other tissues at much lower levels.
Biological function summary

The AR-V7 contributes to the regulation of genes typically involved in cell growth and survival. AR-V7 operates as part of the androgen receptor complex which translocates into the nucleus to influence gene transcription. Due to its persistent activity AR-V7 can drive the expression of androgen-responsive genes which contributes to the proliferation of cells even in low androgen environments. This activity shows why AR-V7 presence creates therapeutic challenges because it promotes unnecessary cell growth.

Pathways

AR-V7 interacts within the androgen signaling pathway which regulates androgen-responsive gene expression. A significant relationship exists between AR-V7 and proteins like coactivators which enhance its transcriptional activity. By bypassing the typical inhibition provided by androgen deprivation therapies (ADTs) AR-V7 sustains pathway activities leading to continued cancer cell survival and proliferation. It is also related to the Akt pathway which further supports cell survival and metabolic processes.

AR-V7 plays an important role in castration-resistant prostate cancer (CRPC) where conventional therapies targeting the androgen receptor fail. The persistent activity of AR-V7 independent of androgen binding results in resistance to ADTs including enzalutamide and abiraterone. AR-V7 expression correlates with poor clinical outcomes in CRPC patients making it a potential biomarker for resistance. Furthermore its interaction with protein elements such as full-length androgen receptors highlights its impact on driving disease progression and resistance to treatments.

Transcription factor activity is modulated by bound coactivator and corepressor proteins like ZBTB7A that recruits NCOR1 and NCOR2 to the androgen response elements/ARE on target genes, negatively regulating androgen receptor signaling and androgen-induced cell proliferation (PubMed : 20812024). Transcription activation is also down-regulated by NR0B2. Activated, but not phosphorylated, by HIPK3 and ZIPK/DAPK3.. Isoform 3. Lacks the C-terminal ligand-binding domain and therefore constitutively regulates the transcription of a specific set of canonical AR-target genes, including PSA/KLK3 and TMPRSS2, independently of steroid hormones (PubMed : 19244107, PubMed : 25008967).
