Biological function summary
NDST3 engages in the modification of heparan sulfate which is an important component of the extracellular matrix. NDST3 is involved in a complex that includes various other NDST family members each contributing distinct functions in heparan sulfate biosynthesis. Through its enzymatic activity NDST3 influences cell signaling and interaction dynamics by producing the variably sulfated domains in heparan sulfate chains that determine ligand binding specificity.
Pathways
NDST3's function in heparan sulfate modification is important in the fibroblast growth factor (FGF) signaling pathway and the Wnt signaling pathway. In these pathways the modified heparan sulfate participates in the regulation of important ligands and receptors which is necessary for proper signal transduction. NDST3 works in concert with proteins such as fibroblast growth factor receptor (FGFR) and Wnt receptors influencing cellular responses and developmental processes.

Essential bifunctional enzyme that catalyzes both the N-deacetylation and the N-sulfation of glucosamine (GlcNAc) of the glycosaminoglycan in heparan sulfate. Modifies the GlcNAc-GlcA disaccharide repeating sugar backbone to make N-sulfated heparosan, a prerequisite substrate for later modifications in heparin biosynthesis. Has high deacetylase activity but low sulfotransferase activity.
