추가 정보

PIGC also known as phosphatidylinositol glycan anchor biosynthesis class C is a protein participating in the biosynthesis of glycosylphosphatidylinositol (GPI) anchors. PIGC functions as an essential component of the GPI transamidase complex. The molecular weight of PIGC is reported to be approximately 34 kDa. It is predominantly expressed in the endoplasmic reticulum but is also present in many tissues throughout the body suggesting its widespread role in various cellular processes.
Biological function summary

PIGC contributes to anchoring proteins to the cell membrane via GPI anchors. It forms part of the multi-subunit enzymatic complex known as the GPI transamidase complex which includes other subunits like PIGA PIGP PIGQ and GPAA1. This complex catalyzes the attachment of newly synthesized proteins to GPI enabling them to embed into membranes which is critical for the functionality of many surface proteins involved in cellular signaling immune response and enzyme activities.

Pathways

A significant aspect of PIGC activity involves its role in the GPI-anchor biosynthesis pathway. This pathway ultimately influences the sorting and surface display of GPI-anchored proteins directly impacting cellular communication and interaction. PIGC's interaction with proteins such as PIGA and PIGQ highlights its integrated action within this pathway and stresses its collaborative function with these proteins to ensure proper cell surface protein anchorage and trafficking.

PIGC has been connected to paroxysmal nocturnal hemoglobinuria (PNH) and certain types of congenital disorders of glycosylation (CDG). PNH is characterized by mutations that affect GPI-anchor biosynthesis leading to hemolytic anemia and other systemic issues. Disorders in the GPI biosynthesis pathway involving proteins like PIGA and PIGQ similarly result in symptoms ranging from cognitive impairments to hypoglycosylation of proteins affirming the critical involvement of PIGC and associated proteins in normal cellular and systemic functions.
