RNF121 also known as Ring Finger Protein 121 is a protein that functions mechanistically as an E3 ubiquitin ligase. This enzyme catalyzes the transfer of ubiquitin from an E2 ubiquitin-conjugating enzyme to a substrate protein tagging it for degradation by the proteasome. RNF121 has a molecular mass of approximately 44 kDa. This protein is widely expressed in the cytoplasm and is abundant in tissues such as the heart brain liver and kidneys amongst others indicating its diverse role in cellular function.
Biological function summary

RNF121 contributes to processes such as protein homeostasis and regulation of signaling pathways through its involvement in the ubiquitin-proteasome system. It does not function alone; RNF121 often associates with other cellular proteins to form complexes that target specific proteins for ubiquitination. This process is integral for controlling the degradation of damaged or misfolded proteins and for regulating diverse signaling pathways within the cell.

Pathways

The ubiquitin ligase activity of RNF121 plays roles in the Wnt signaling pathway and endoplasmic reticulum-associated degradation (ERAD) pathway. In the Wnt pathway RNF121 interacts with proteins like beta-catenin modulating its stability and consequently influencing gene expression related to cell growth and differentiation. In the ERAD pathway RNF121 helps manage protein quality control by targeting misfolded proteins for degradation ensuring cellular health and function.

RNF121 has been linked to cancer and neurodegenerative diseases. Mutations or dysregulation of RNF121 can influence the stability of key regulatory proteins leading to unregulated cellular proliferation associated with tumor progression. Similarly alterations in RNF121 might disrupt protein degradation processes contributing to the accumulation of misfolded proteins observed in neurodegenerative conditions. Connections between RNF121 and proteins involved in these diseases such as p53 in cancer or tau in Alzheimer’s disease highlight its role in disease mechanisms.
