SAMSN1 also known as HACS1 or SLY2 is a protein that functions primarily as an adapter protein involved in signal transduction. It has a mass of approximately 46 kDa. This protein is expressed in several tissues with high levels found in lymphoid organs like the spleen and peripheral blood leukocytes. SAMSN1 modulates protein-protein interactions through its SAM (sterile alpha motif) domain and a SH3 (Src homology 3) domain enabling it to interact with other critical proteins within cells.
Biological function summary

The function of SAMSN1 is essential in the immune response and cell signaling processes. It is part of signaling complexes that regulate immune cell function particularly B cells and T cells. By binding to other signaling proteins SAMSN1 can influence pathways that control cell growth differentiation and survival. This protein also participates in modulating transcription factors that are critical for immune cell maturation and function.

Pathways

SAMSN1 is significantly involved in the B-cell receptor signaling and the PI3K/Akt pathway. Through these pathways SAMSN1 affects the regulation of signal transduction cascades involved in cell proliferation and survival. It interacts with proteins such as PI3K and Akt which are significant in transducing signals that impact cellular responses to external stimuli.

SAMSN1 has been linked to autoimmune diseases and cancer. Altered expression of this protein can contribute to the development of conditions such as systemic lupus erythematosus and certain lymphomas. Interactions between SAMSN1 and proteins like BCR (B-cell receptor) play a role in the pathology of these disorders where dysregulation in signaling pathways may lead to the abnormal proliferation of immune cells or their improper functioning.
