Statherin also known as STA-1 is a small protein with a molecular mass of roughly 5.4 kDa. This protein is found predominantly in human saliva secreted by the salivary glands including the parotid gland. Statherin plays a mechanical role in maintaining the stability of the mineral content in saliva which is important for the prevention of calcium phosphate precipitation. By binding calcium ions Statherin keeps the saliva supersaturated with calcium phosphate which helps prevent unwanted crystallization and maintains the fluidic integrity of the oral cavity.
Biological function summary

Statherin’s function is essential for oral health by playing a significant part in the protection of tooth enamel. It inhibits the spontaneous nucleation of calcium phosphate salts essentially preventing their accumulation on teeth. Its ability to bind to the tooth surface also forms a part of the pellicle a protein film that covers the enamel. Statherin works as a solitary protein in this context and does not usually form complexes with other biomolecules in saliva. However its protective impact on tooth surfaces directly influences oral health by maintaining a balanced enamel surface.

Pathways

Statherin is an important element in the homeostasis of mineralized tissues specifically within the calcium precipitation pathway. It relates to molecules that modulate calcium levels involving interactions with other phospho-proteins such as proline-rich proteins and histatins which also contribute to mineral balance and oral health. These pathways underline the importance of Statherin as a component of the oral environment's stability and function.

Statherin associates closely with dental calculus and caries. Its ability to inhibit calcium phosphate precipitation directly protects against dental plaque formation which if left uncontrolled leads to calculus. Furthermore caries development can relate to inadequate Statherin function highlighting its significance in preventive dentistry. Interactions with proteins like proline-rich proteins in the formation of the enamel pellicle are critical as they can diminish the likelihood of tooth decay by controlling bacterial colonization and mineral deposition.

일반 정보

기능

Salivary protein that stabilizes saliva supersaturated with calcium salts by inhibiting the precipitation of calcium phosphate salts. It also modulates hydroxyapatite crystal formation on the tooth surface.

서열 유사성

Belongs to the histatin/statherin family.

Post-translational modifications

Substrate for transglutaminase-2. More than 95% of the cyclized peptide is cyclo-statherin Q-37, and less than 5% is cyclo-statherin Q-39. Cyclized forms account for about 1% of total statherin in saliva.. Sulfated on tyrosine residues.
