STEAP2 also known as six transmembrane epithelial antigen of the prostate 2 is a metalloreductase with a mass of about 56 kDa. It facilitates the reduction of metal ions in a process dependent on FAD and NADPH. Expressed mainly in the prostate STEAP2 also appears in other tissues at lower levels. This protein exhibits a tendency to localize to cellular membranes and endocytic vesicles aiding in the uptake and processing of metal ions like iron and copper which are important for cellular activities.
Biological function summary

STEAP2 plays a role in ion homeostasis and cellular proliferation. It acts as part of a complex involving other STEAP family members contributing to the reduction of metal ions essential for various cellular processes. Its ability to mediate electron transport is important for maintaining the balance of redox reactions critical for normal cellular functions. The involvement in cellular proliferation ties STEAP2 to tissue development and repair mechanisms.

Pathways

STEAP2 interacts within metal ion transport and homeostasis pathways influencing processes like oxidative stress responses. It associates with proteins such as ferritin important for iron storage and metallothioneins linked to metal detoxification. These interactions are essential for regulating intracellular metal ion concentrations thereby impacting cellular health and function. It also relates to cellular growth pathways connecting it to certain growth factor signals.

Abnormal STEAP2 expression links to prostate cancer and other proliferative disorders. Its overexpression in prostate cancer cells correlates with increased tumor aggressiveness and progression. STEAP2 interacts with other oncogenic proteins such as PSA (prostate-specific antigen) which are critical in the diagnosis and progression monitoring of prostate cancer. Additionally alterations in STEAP2 function may affect systemic metal ion homeostasis potentially contributing to disorders like hemochromatosis although this link is less well-defined in clinical settings.

일반 정보

기능

Integral membrane protein that functions as a NADPH-dependent ferric-chelate reductase, using NADPH from one side of the membrane to reduce a Fe(3+) chelate that is bound on the other side of the membrane (By similarity). Mediates sequential transmembrane electron transfer from NADPH to FAD and onto heme, and finally to the Fe(3+) chelate (By similarity). Can also reduce Cu(2+) to Cu(1+) (By similarity).

서열 유사성

Belongs to the STEAP family.
