Biological function summary
TAF10 acts as an integral part of the transcription factor IID (TFIID) complex which is essential for RNA polymerase II-driven transcription initiation. TAF10 contributes to the recognition and binding of promoter DNA sequences facilitating transcription initiation. It also serves a regulatory role in mediating interactions between the TFIID complex and other transcription factors enhancing transcriptional control and efficiency.

TAF10 plays an important role in the regulation of gene expression and RNA polymerase II transcription pathways. It interacts with various components of the transcription initiation complex to initiate and regulate transcription. TAF10 associates with other TAFs and TBP (TATA-binding protein) in the TFIID complex together forming a scaffold for the correct assembly of RNA polymerase II.

The TFIID basal transcription factor complex plays a major role in the initiation of RNA polymerase II (Pol II)-dependent transcription (PubMed : 33795473). TFIID recognizes and binds promoters with or without a TATA box via its subunit TBP, a TATA-box-binding protein, and promotes assembly of the pre-initiation complex (PIC) (PubMed : 33795473). The TFIID complex consists of TBP and TBP-associated factors (TAFs), including TAF1, TAF2, TAF3, TAF4, TAF5, TAF6, TAF7, TAF8, TAF9, TAF10, TAF11, TAF12 and TAF13 (PubMed : 33795473). TAF10 is also component of the PCAF histone acetylase complex, the TATA-binding protein-free TAF complex (TFTC) and the STAGA transcription coactivator-HAT complex (PubMed : 10373431, PubMed : 11564863, PubMed : 12601814, PubMed : 18206972, PubMed : 9885574). May regulate cyclin E expression (By similarity).

Post-translational modifications
Monomethylated at Lys-189 by SETD7, leading to increased affinity for RNA polymerase II.. Lysine deamination at Lys-189 to form allysine is mediated by LOXL2. Allysine formation by LOXL2 results in release of TAF10 from promoters, leading to inhibition of TFIID-dependent transcription.
