Thyrotropin Releasing Hormone (TRH) also known as thyroliberin is a tripeptide hormone with a molecular mass of approximately 362.4 Da. This hormone mainly expresses in the hypothalamus of the brain. TRH plays an important role in the regulation of the thyroid axis by stimulating the secretion of thyroid-stimulating hormone (TSH) from the anterior pituitary gland. Additionally it influences the release of prolactin another pituitary hormone.
Biological function summary

TRH serves important regulatory functions within the endocrine system. Notably TRH is an initiator in the hypothalamic-pituitary-thyroid (HPT) axis. It does not form part of a larger complex but acts as a standalone signaling molecule. This hormone facilitates communication between the central nervous system and the endocrine system influencing metabolic processes and affecting growth and development via its role in TSH and prolactin secretion.

Pathways

TRH activates specific cell surface receptors linked to G-protein coupled signaling which increases the production of intracellular second messengers such as inositol triphosphate (IP3) and calcium ions. One significant pathway is the HPT axis which integrates TRH into a feedback cycle with thyroid hormones like thyroxine (T4) and triiodothyronine (T3). Proteins related to TRH within this signaling pathway include its target TSH which in turn regulates the biosynthesis of T3 and T4 in the thyroid gland.

TRH has been implicated in conditions such as hypothyroidism and major depressive disorder.
