Transducin-like enhancer of split 4 (TLE4) is a transcriptional corepressor involved in the regulation of gene expression. It is also known as ESG (Enhancer of split groucho) or GRG4. TLE4 belongs to the Groucho/TLE protein family which shares a general structure characterized by WD40 repeats. The protein has a molecular mass of about 82 kDa. TLE4 expresses in several tissues including the brain heart and peripheral blood leukocytes indicating its broad functional importance.
Biological function summary

TLE4 participates in the repression of transcription by forming a multiprotein complex. It acts as a corepressor that interacts with various transcription factors often modulating the expression of genes involved in developmental processes. TLE4 commonly partners with transcription factors from the basic helix-loop-helix (bHLH) family which play a significant role in the regulation of neuronal and muscular development. The repression activity of TLE4 makes it a temporal regulator helping maintain and coordinate gene expression during cellular differentiation.
