Biological function summary
TLE4 participates in the repression of transcription by forming a multiprotein complex. It acts as a corepressor that interacts with various transcription factors often modulating the expression of genes involved in developmental processes. TLE4 commonly partners with transcription factors from the basic helix-loop-helix (bHLH) family which play a significant role in the regulation of neuronal and muscular development. The repression activity of TLE4 makes it a temporal regulator helping maintain and coordinate gene expression during cellular differentiation.
Pathways
TLE4 functions within the Wnt signaling and Notch signaling pathways. Both of these pathways are important for cell fate determination and organ development. In the Wnt pathway TLE4 interacts with beta-catenin to modulate transcriptional activity contributing to cellular responses to external signals. In the Notch signaling pathway TLE4 associates with core transcription factors like CSL (CBF1/Suppressor of Hairless/Lag1) to regulate the expression of Notch target genes facilitating diverse developmental processes.

Transcriptional corepressor that binds to a number of transcription factors. Inhibits the transcriptional activation mediated by PAX5, and by CTNNB1 and TCF family members in Wnt signaling. The effects of full-length TLE family members may be modulated by association with dominant-negative AES. Essential for the transcriptional repressor activity of SIX3 during retina and lens development and for SIX3 transcriptional auto-repression (By similarity). Involved in transcriptional repression of GNRHR and enhances MSX1-mediated transcriptional repression of CGA/alpha-GSU (By similarity).
