Tau-tubulin kinase 1 (TTBK1) also known as TTBK-1 plays an important role in phosphorylating proteins involved in microtubule dynamics. It has a molecular weight of approximately 150 kDa. TTBK1 is highly expressed in the central nervous system especially in the brain but also occurs in other tissues to a lesser extent. The protein functions as a serine/threonine kinase targeting tau and tubulin proteins amongst others which are essential for maintaining neuronal structure and function.
Biological function summary

This kinase takes part in stabilizing microtubules by phosphorylating tau a protein important for microtubule assembly and stabilization. TTBK1's action helps regulate neuronal cell signaling and maintains proper axonal transport. It does not function in solitude but associates with other proteins as it may form part of larger multi-protein complexes essential for executing its functions. The phosphorylation activity of TTBK1 modulates interactions between microtubules and other cytoskeletal elements in neurons.

Pathways

TTBK1 activities are integral to the tau phosphorylation pathway and the Wnt signaling pathway. In the tau phosphorylation pathway it works alongside other kinases like glycogen synthase kinase 3 beta (GSK3β) contributing to tau modifications affecting microtubule stability. Moreover through Wnt signaling TTBK1 influences pathways responsible for synaptic plasticity and nerve cell communication interacting with proteins like beta-catenin and the Dishevelled protein family. These pathways impact neuron development and functioning.

TTBK1's role is linked significantly to Alzheimer's disease and other neurodegenerative disorders. The over-phosphorylation of tau by TTBK1 contributes to tau aggregation forming neurofibrillary tangles a hallmark of Alzheimer's disease. In this context TTBK1 interacts with tau and partners with GSK3β in pathways that lead to abnormal tau phosphorylation. Mutations or dysregulation of TTBK1 could also contribute to Parkinson's disease emphasizing its involvement in neurodegeneration processes driven by improper tau metabolism and/or interaction with alpha-synuclein.
